Combined L-citrulline and glutathione supplementation increases the concentration of markers indicative of nitric oxide synthesis

Abstract

Background

Nitric oxide (NO) is endogenously synthesized from L-arginine and L-citrulline. Due to its effects on nitric oxide synthase (NOS), reduced glutathione (GSH) may protect against the oxidative reduction of NO. The present study determined the effectiveness of L-citrulline and/or GSH on markers indicative of NO synthesis in in vivo conditions with rodents and humans and also in an in vitro condition.

Methods

In phase one, human umbilical vein endothelial cells (HUVECs) were treated with either 0.3 mM L-citrulline, 1 mM GSH (Setria®) or a combination of each at 0.3 mM. In phase two, Sprague–Dawley rats (8 weeks old) were randomly assigned to 3 groups and received either purified water, L-citrulline (500 mg/kg/day), or a combination of L-citrulline (500 mg/kg/day) and GSH (50 mg/kg/day) by oral gavage for 3 days. Blood samples were collected and plasma NOx (nitrite + nitrate) assessed. In phase three, resistance-trained males were randomly assigned to orally ingest either cellulose placebo (2.52 g/day), L-citrulline (2 g/day), GSH (1 g/day), or L-citrulline (2 g/day) + GSH (200 mg/day) for 7 days, and then perform a resistance exercise session involving 3 sets of 10-RM involving the elbow flexors. Venous blood was obtained and used to assess plasma cGMP, nitrite, and NOx.

Results

In phase one, nitrite levels in cells treated with L-citrulline and GSH were significantly greater than control (p < 0.05). In phase two, plasma NOx with L-citrulline + GSH was significantly greater than control and L-citrulline (p < 0.05). In phase three, plasma cGMP was increased, but not significantly (p > 0.05). However, nitrite and NOx for L-citrulline + GSH were significantly greater at 30 min post-exercise when compared to placebo (p < 0.05).

Conclusions

Combining L-citrulline with GSH augments increases in nitrite and NOx levels during in vitro and in vivo conditions.

Doctor's Best NAC Detox Regulators with Seleno Excell, Non-GMO, Vegetarian, Gluten Free, Soy Free

From the manufacturer:

NAC supports numerous detox functions NAC's detox value is grounded in its rapid conversion to cysteine that protects the blood and body fluids; the cells' need for cysteine to make glutathione; and direct antitoxin effects by NAC itself.

Supplies cysteine to make glutathione. Taken by mouth, NAC elevates cysteine in the blood. Cysteine is the predominant antitoxin and antioxidant of the blood and other body fluids.

Taking NAC is actually a better way to get cysteine than cysteine itself (l-cysteine), which is too unstable to be used in dietary supplements. NAC's acetyl group keeps it stable for its absorption into the blood, then subsequently its acetyl group is removed (mostly by liver enzymes) and the molecule becomes cysteine.

Cysteine may be a conditionally essential nutrient. The diet of some human populations may be relatively deficient in cysteine, and (beginning around the fourth decade of life) glutathione levels decline with age.

A pioneering cysteine researcher (w. Droge) strongly advocated taking NAC to support healthy aging, suggesting "everybody is likely to experience a cysteine deficiency sooner or later." Sustains the body's glutathione status.

Glutathione is the most highly concentrated antitoxin and antioxidant within human cells, and adequate glutathione is a must for good health. Glutathione is used to neutralize toxins, whether these originate outside the body or are naturally produced by the body. Healthy cells continually "top up" their glutathione stores by importing cysteine from the blood (not glutathione), then making new glutathione from scratch.

The supply of cysteine is normally the limiting factor for the cells to make glutathione. Supplementing with NAC therefore helps sustain both cysteine levels in the blood, and glutathione levels inside the cells.

Finally, Someone with the Huevos to make a real pre-workout. Pre-Jym by Jim Stoppani | It's not a game!

Pre Jym is the first pre-workout supplement designed by Jim Stoppani, a personal nutrition and health consultant for celebrity clients like The Rock and LL Cool J.

If you don't know who Jim Stoppani is then my question is ..... do you even lift?

Currently pre-workouts on all only energy drinks that have no significant amounts of ingredients.

Well, building muscle is not a game and you should be getting the full amount of key ingredients to make you grow.

Though an official supplement label for Pre Jym hasn’t been released yet, the following ingredients are featured on the Pre Jym advertisements on Stoppani’s Facebook page.

BCAAs (6 g)
Branched-chain amino acids, or BCAAs, are considered the “building blocks of muscle.” They make up 35 percent of muscle mass and are considered essential for healthy muscular growth and development. BCAAs stimulate insulin production, which means muscle cells can draw from circulating blood sugar as an energy source [1].

Citrulline Malate (6 g)
L-citrulline is an amino acid used for improving focus as well as improving muscle strength. Incidentally, it also prevents heart disease.
Citrulline malate is a form especially adapted to enhancing anaerobic exercise, making it effective in products like Pre Jym. A 2010 study on citrulline malate showed an increase in pectoral reps and sets, as well as a 40% decrease in muscle soreness [2].

Creatine HCL (2 g)
A new creatine form, creatine HCL was created in 2003 when researchers attempted to synthesize creatine ethyl ester ingredients. A highly concentrated creating monohydrate form with a hydrochloride group attached, creatine HCL has unique properties.
Like creatine monohydrate, creatine HCL boosts muscle growth and stimulates insulin-growth factor 1 (IGF-1) production. However, it causes fewer side effects because it is more water soluble. In one study, researchers found it to be 59 times more water soluble than creatine monohydrate [3].

Beta-Alanine (2 g)
Beta-alanine is responsible for the “tingle” you feel with pre-workout formulas like Pre Jym. Said to improve athletic performance and exercise capacity, beta-alanine builds lean muscle mass and improves flexibility. It is said to be particularly effective alongside creatine [4].

Betaine (1.5 g)
Betaine hydrochloride is a synthetic chemical said to enhance energy and boost potassium levels. This supports a longer, more productive workout with a healthier outcome. It is unknown exactly how betaine hydrochloride works, however.

N-Aceyl L-Cysteine (NAC) (600 mg)
NAC is a modified cysteine molecule. Like oxo-proline, it has a high antioxidant count. These antioxidants boost the immune system and improve health, but do nothing to raise HGH levels. However, when NAC is metabolized, it produces glutathione. Glutathione in turn increases DHEA and IGF-1 levels, which promote greater HGH release [5].

Alpha GPC (150 mg)
Alpha GPC is a natural choline compound found in the brain and in milk. Alpha-GPC influences growth hormone production by stimulating the pituitary gland to release more HGH. This influences lean muscle mass growth and improves mental performance during workouts. Alpha GPC also increases speed and agility [6].

Caffeine (300 mg)
Caffeine is often used in diet pills to increase metabolism and burn fat. In a muscle-building supplement like Pre Jym, those effects improve your body composition. However, caffeine also improves exercise performance by increasing energy levels.

In a 1978 study, caffeine boosted exercise performance in members of a cycling group [7]. Those who took caffeine averaged 90.2 minutes of continuous cycling, while subjects in a control group managed just 75.5 minutes.

Huperzine (50 mcg)
Huperzine is derived from Chinese blub moss. Used to boost mental focus, huperzine is believed to prevent Alzheimer’s disease. This helps you concentrate on your workout and get better results [8].

What Flavors Are Available?

According to Jim Stoppani’s Twitter account, Pre Jym Flavors include:
• Watermelon
• Fruit Punch

The Power of Pre JYM
No Proprietary Blends
Look at the supplement facts panel on your favorite pre-workout product and you'll most likely see the term "proprietary blend." Supplement companies want you to think this means they've created a special blend of ingredients that magically work together to deliver unique results. Because this blend is so amazing, they have to keep the doses of each ingredient secret so no other company steals the formula.
The truth, however, is that propriety blends serve a different purpose. They allow supplement companies to hide how little of each ingredient they've actually put in a product. This results in cheap, underdosed, ineffective supplements.
Pre JYM doesn't use proprietary blends. Take a look at its supplement facts pane and you'll see the actual doses of all 13 ingredients. I have nothing to hide. I'm proud of the amount of each and every ingredient I've included. Together, those 13 ingredients in those exact doses are the closest thing to a magical formula that you're going to find.
Proper Doses
For the reasons detailed above, many supplement companies are moving toward transparency and axing proprietary blends. That's a good thing. However, these companies are still guilty of grossly underdosing ingredients.
Take citrulline malate, for example. The research on this potent amino acid shows that you need 6 grams per dose. Most pre-workout products leave out citrulline entirely, and none of them provide anywhere close to 6 grams. Until now.

No "Abbreviated" Formulas
A handful of companies are as fed up as I am with proprietary blends and underdosed formulas, but they fall short by using "abbreviated" formulas. An abbreviated formula is one that uses proper dosing of all ingredients, but only includes a few ingredients.
Take, for example, a pre-workout formula that includes beta-alanine, creatine, caffeine, and tyrosine. There's nothing wrong with it, but it could be made a lot more effective through the addition of ingredients like citrulline malate, betaine, and BCAAs, to name just a few. Why would companies not create the most robust formula possible? Because adding more ingredients costs too much money.
Pre JYM includes 13 ingredients that work in concert to prime the body for intense workouts, making it the most complete pre-workout supplement available.*
No Concentrates
Many companies also try to trick buyers by calling their pre-workout product "concentrated." A serving of one of these "concentrated" products can contain as few as 3 to 6 grams of powder. What kind of magic did they use to cram enough creatine, beta-alanine, citrulline, arginine, caffeine, and other ingredients into that tiny dose? They didn't use any magic, which is why all of those "concentrated" formulas also include proprietary blends. That's also why the serving size of Pre JYM is more than 26 grams. It contains 13 ingredients at proper, powerful doses.
JS, Not BS
I've been studying supplement science in the lab and gym for decades. I've been delivering supplement advice to millions of people in the pages of Muscle & Fitness, FLEX, and Muscle & Fitness Hers, as well as here on Bodybuilding.com and my own website, JimStoppani.com, for over a decade.
Most importantly, I've been training since I was 11 years old. Before each of my workouts, I would draw on all of my experience and all of my education to concoct my own pre-workout supplement, combining ingredients from 13 different bottles and tubs.
That's why I developed Pre JYM. I wanted to give my friends, colleagues, all of you, and myself the ability to access, in one bottle, the best ingredients in the exact doses suggested to help build lean muscle, torch body fat, improve strength, and boost speed.*
Prepare to experience the power of real science, real ingredients, and unreal results.

ADVANCED CYCLE SUPPORT Rx™by Iron Mag Labs not just for juicers and gear guys.

The ingredients in this concoction are just plain great for the body.

This product contains 7 key ingredients for a complete 'on cycle' protection and support. Per 2 Capsules.

1. Milk Thistle (Silybum marianum) 500MG - One function of the liver is to detoxify and remove toxins, including heavy metals and chemotherapy, from the body. Milk thistle is a liver protective. It is an antioxidant that helps repair damaged liver cells. Studies have shown evidence that silymarin acts almost solely on the liver and kidney. The antioxidant properties in silymarin are believed to be the protective factors. Laboratory studies demonstrate that silymarin functions as a potent antioxidant, stabilizes cellular membranes, stimulates detoxification pathways, stimulates regeneration of liver tissue, inhibits the growth of certain cancer cell lines, exerts direct cytotoxic activity toward certain cancer cell lines.

2. NAC (N-Acetylcysteine) 400MG - NAC can help to prevent damage to the liver being a powerful anti-oxidant and cell detoxification co-factor, NAC works to eliminate your body of free radicals and heavy metals. N-Acetyl-Cysteine is currently the dietary supplement of choice for building up cysteine or conserving the body's store of Glutathione, Cysteine and other Sulfhydryl anti-oxidant resources. This is very crucial for the body's life functions, as NAC helps the body neutralize toxins, heavy metals, such as mercury.

3. Hawthorne Berry 108% extract 300MG- is highly recognized herb for the heart and cardiovascular system. It has the ability to dilate (enlarge or open) the coronary arteries (the vessels that supply blood to the heart), thus improving blood and oxygen supply to the heart muscle. It also strengthens the hearts pumping ability (muscle), helping the heart to beat more forcefully and efficiently. In addition, hawthorne appears to dilate the highways of other blood vessels around the body, thereby allowing blood to circulate more freely with less strain on the heart. Another benefit of hawthorne berry is that it harbors potent antioxidant properties, which are believed to exert cholesterol lowering effects and reduce the accumulation of fatty plague in the arteries – the hallmark of atherosclerosis.

4. Saw Palmetto (Serenoa repens) 45% extract 200MG - is used popularly in Europe for symptoms associated with benign prostatic hypertrophy (enlargement of the prostate). Although not considered standard of care in the United States, it is the most popular herbal treatment for this condition. Saw palmetto was listed in the United States Pharmacopeia from 1906 to 1917 and in the National Formulary from 1926 to 1950. Saw palmetto extract is a licensed product in several European countries. Multiple mechanisms of action have been proposed, and saw palmetto appears to possess 5-?-reductase inhibitory activity (thereby preventing the conversion of testosterone to dihydrotestosterone). Hormonal/estrogenic effects have also been reported, as well as direct inhibitory effects on androgen receptors and anti-inflammatory properties.

4. Coenzyme Q10 (CoQ10) 100MG - boosts energy, enhances the immune system, and acts as an antioxidant. Clinical research suggests that using coenzyme Q10 supplements alone or in combination with other drug therapies and nutritional supplements may help prevent or treat some of the following conditions: Heart disease, High blood pressure and High cholesterol. Researchers believe that the beneficial effect of CoQ10 in the prevention and treatment of heart disease is due to its ability to improve energy production in cells, inhibit blood clot formation, and act as an antioxidant. Several clinical studies involving small numbers of people suggest that CoQ10 may lower blood pressure. Levels of CoQ10 tend to be lower in people with high cholesterol compared to healthy individuals of the same age.

5. Celery Seed Extract 6:1 concentrate 75MG- 3nB is the active compound that is unique to celery. 3nB was discovered as the active component of celery in response to investigations by researchers seeking to explain some of the medicinal effects of celery including the lowering of blood pressure and the relief of arthritis. High blood pressure is usually the result of too much fluid there is in your blood and how flexible or resistant your blood vessels are. Retention of sodium (salt) leads to increase fluid volume in the blood while hardening of the arteries and the hormones released during stress lead to loss of flexibility or constriction of blood flow. In treating high blood pressure, doctors usually prescribed diuretics (water pills) to reduce the fluid volume and vasodilators to relax the arteries to reduce the resistance of blood flow or beta-blockers to turn down the pumping action of the heart. 3nB appears to help lower blood pressure by both acting as a diuretic and vasodilator through impacting the production of prostaglandins as well as acting in a similar manner to drugs known as calcium-channel blockers. 3nB has also been shown to lower blood cholesterol levels and reduce the formation of arterial plaque in experimental studies. This effect may increase the elasticity of the blood vessels and also lead to lower blood pressure readings. 3nB also appears to promote some effects on areas and systems of the brain that control vascular resistance.

6. Grape Seed Extract 95% extract 75MG- is a natural plant substance that has a concentrated source of oligomeric proanthocyanidins (OPC). These anti-oxidants help protect cells from free radical damage and also promotes healthy circulation. Grape Seed Extract is rich in polyphenols, a compound that's high in antioxidants. Studies have shown OPC to be more powerful antioxidants than vitamin C, E, and beta-carotene. There are countless studies that demonstrates the many health benefits of grape seed extract. It has been extensively researched across the globe. In the research of Dr. Jacques Masquelier et al., the Pasteur & Huntington Institutes and 7 other leading Universities in Europe, Grape Seed Extract has been shown exceptionally effective fighting against free radicals in the body.
